Negative-staining technique was used to investigate whether the
extract ZG from Gardenia could change or destroy the virus
envelope protein structure to weaken HSV-1 virulence.After treated with
extract ZG from Gardenia,the HSV-1 particles and the virus attachment proteins(VAPs) on virus surface were observed under electron microscope.Fluorescein isothiocyanate(FITC) as HSV-1 fluorescent probe and heparin sodium as the control sample,we adopted the
Cooling Charged-Coupled Device(Cccd) and Aquacomos analysis software to study the effect of extract ZG from Gardenia on the cell adsorption quantity of HSV-1 in different methods.The results showed that extract ZG from Gardenia didn′t directly destroy VAPs of HSV-1.Both the group adding heparin sodium first and the group adding heparin sodium and virus simultaneously could prevent the viral binding to Hep-2 cells.Adding extract ZG from Gardenia in different ways all could prevent the HSV-1 binding to the Hep2 cell surface and reduce the virus adsorption quantity.
