Objective:To research the gene expressions of injury on blood vessel and
vascular endothelial cell apoptosis induced by mannitol
in using vitamin C.Methods:(1) Animal experiment: healthy rabbits were randomized into 4 groups.Group A was injected with mannitol into the vein of the rabbit ear.Group B,group C,and group D,were injected first with NS,VitC and VitE respectively to pretreat the vessel,and then with mannitol.Electronic lens and optical microscope were used to observe the injury of blood vessel.(2) Arrays of type Biostar H40s were used to test the changes of apoptosis gene expressions induced by mannitol before and after using protective agents.Results:(1) As for the degree of injury of the blood vessel,group A was serious,group B was moderate to serious,and group D is slight.No pathological changes were observed in Group C.(2) All 330 apoptosis-related genes: in chips
pretreated by normal saline,none was significantly different.In chips pretreated by vitamin C,45 apoptosis genes were down-regulated,34 anti-apoptosis genes were up-regulated.In chips pretreated by vitamin E,only 3 apoptosis genes were down-regulated.Conclusions:Vitamin C can protect
vascular endothelial cell.
