Influence of celebrex and mizolastine on rat paw edema models

Objective:To investigate the anti-inflammatory properties of celebrex, mizolastine and explore their possible mechanisms
of anti-inflammation. Methods: The inflammatory edema reaction of rat paw was induced by a single subcutaneous injection of arachidonic acid (AA) or carrageenan. The tested drugs (celebrex, mizolastine, dexamethasone and Tween 80) were given orally 2 hours before the injection of above inflammatory mediators. Rat paw volume was measured plethysmographically at 1, 2, 3, 4h after the mediator injections and the inhibitory effects were determined by the changes of rat paw volumes. Results: Arachidonic acid or carrageenan could induce intensive and persistent paw edema. The intensities of carageenan-induced edema were selectively inhibited by celebres significantly (P<0.05). Conclusion: Celebrex possesses anti-inflammatory properties via inhibition of cyclooxygenases, while the anti-inflammatory effect of mizolastine is related to the inhibition of the 5- lipogenase pathway. Combination of celebrex with mizolastine is suggested to be more effective in controlling inflammatory reaction of allergic diseases.
