AIM : To study the effects and mechanisms of rosiglitazone and gliclazide on
insulin resistance in rats induced by high
fat diet and hyperlipemia. METHODS : After seven weeks feeding with high fat diet, the rats with
insulin resistance were divided into 3 groups: the control group, the rosiglitazone group and the gliclazide group. Insulin and glucose tolerance were tested.
Concentrations of tumor necrosis factor α (TNF α) and fasting insulin (Fins) in serum and triacylglycerol (TG), malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione (GSH) were determined in hepatic. RESULTS : The state of impaired glucose tolerance (IGT) was improved, the concentrations of fasting glucose (FSG), TNF α, TG and MDA were decreased ( BX P < 0.05 ), and the deposit of GSH were increased ( P < 0.01 ) by rosiglitazone and gliclazide. The high insulin level and the concentrations of blood (BUN), creatine (Cr) and Fins were decreased and the sensitivity of insulin (ISI) ( P < 0.05 ) was increased by rosiglitazone. CONCLUSION : Rosiglitazone can improve the insulin resistance in rats induced by high fat diet and hyperlipemia.
