Objective: To compare the eye penetration of ofloxacin on intravenous and topical way after to determine the concentration
and dynamic changes of ofloxacin in aqueous humor and vitreous humor. Methods: Forty-eight Japanese big ear white rabbits were randomly divided into twelve groups. The aqueous humor and vitreous humor were collected at 15,30,45,60andl20minutes following intravenous injection of 10mg/kg of ofloxacin in 5 groups and topical tarivid drop every 15 minutes for 5 times in another 5 groups.The rest groups are blank ones.All concentration of ofloxacin was determined by the RP-HPLC. Results: Aqueous humor concentration of ofloxacin was significantly highter than vitreous humor of that at the same period. Nothing was found in the vitreous humor of topical ofloxacin groups. On the other side, ofloxacin was found in the vitreous humor of intravenous ofloxacin groups. Conclusion: Ofloxacin has the good eye penetration by intravenous way that can pass through the blood-retinal barrier, which suggests intravenous ofloxacin could be the first choice in the prevention and treatment of endophthalmitis.
