OBJECTIVE To study the
pharmacokinetics and relative
bioavailability of the nicotinic ac id sustained-release capsuless in dogs.METHODS 6 dogs were divided into two groups by double crossing method in two weeks.The concentrations of
nicotinic acid in plasma were determined by ion-paired revers ed-phase HPLC. The relative bioavailability and pharmacokinetic parameters of n icotinic acid sustained-release
capsules were computed by 3P97and compared wit h those of nicotinic acid
tablets in the market. RESULTS The pharmacokinetics of nicotinic acid sustained-relea se capsules and nicotinic acid tablets in the market were fitted to one-compart ment modle.The pharmacokinetic parameters of sustained release capsules and imm ediate release tablets were T max ( 2.9± 1.6) and ( 0.9± 0. 6)h;C max ( 5.7± 1.6) and ( 11.7± 5.8) mg·L -1;AUC 0-t ( 24.8± 13.8) and ( 22.4± 10.6) mg·L -1·h respectively,The relative
bioavilability of the sustained rele ase capsules was 110.58% . CONCLUSION
nicotinic acid sustained-release capsules have mark ed sustained-release characters.
