AIM To study the effect of the combination of
promoting hepatocyte growth factor and insulin on pancreatic islet secretry
function of diabetic mice.METHODS Seventy-five diabetic mice induced by streptozotocin were divided into 5 groups,fifteen diabetic mice each group.Those groups were separately treated by insulin(2 U·kg -1,im),
nicotinamide(1 g·kg -1,ig),
promoting hepatocyte growth factor(20 mg·kg -1,iv),promoting hepatocyte growth factor(20 mg·kg -1,iv) combined with insulin(2 U·kg -1,im),NS(same volume,1 mL,iv).Three months later,blood insulin and C-peptide were analysed by radioimmunoassay.Pathological photos were made by immunohistochemical staining and were observed.β cell numbers in them were counted.RESULTS Compared with NS, the combination of promoting hepatocyte growth factor and insulin could significantly improve blood insulin and C-peptide concentrations(P<0.01).Islet β cells were significantly increased. Big or small β cells were observed by microscope.CONCLUSION The combination of promoting hepatocyte growth factor and insulin could significantly improve pancreatic secretry function and have the effect of regenerating β cell.
