URPOSE To study the effect of orallyadministered earthworm fibrinolytic enzyme (EFE) on the activity of plasmin and platelet
aggregation.METHODS The healthy Beagles dogs (6.5~9kg) were orally administered with a single/multiple 40 or 80mg/kg dose of EFE, and venous blood was drawn for determination. The activity of plasmin was determined by means of chromogenic substrate S-2251. ADP 50μmol/L was used as an inducer to measure the platelet aggregation.RESULTS (1) After the first dose of EFE administration, the activity of plasmin of Beagles dogs was increased in a dosedependent manner (P<0.05). Similar increase was observed with successive oral administration for 10d, but there was no significant difference between the 40mg/kg and 80mg/kg groups. (2) The aggregation of platelet was inhibited by EFE 40mg/kg and aspirin 8mg/kg 3h after administration (P<0.05). No significant effect was found while EFE 80mg/kg and amylum was administered orally.CONCLUSIONS EFE increases the activity of plasmin and inhibits platelet aggregation in normal Beagles dogs after oral administration.
