AIM:To observe the influence on conversion of myofibroblasts(MFB) from hepatic stellate cells(HSC) in vitro by drug serum
of fufang biejia ruangan fang,and to explore its mechanism in preventing the hepatic fibrosis. METHODS:Seventy five adult healthy male SD rats were randomly divided into normal control group, model group,high,middle and low dose groups(fufang biejia ruangan fang),and the serum was prepared in all the groups.The HSCs were separated with the HSC separating technique,and then cultured in different serum for 24 and 48 hours,then the expression of alpha smooth muscle actin(α SMA) in HSC was detected with immunohistochemistry ABC,and then the quantitative analysis was performed by image analysis system. RESULTS:The absorbance(A) of the serum to the different α SMA positive staining in HSC in the high,middle and low dose drug groups at 24 hours(110.95±45.21,243.82±62.97 and 417.46±126.72) and 48 hours(333.71±57.90,345.08±51.85 and 675.35±202.20) were significantly different from those in the model group<778.93±190.26 and 1 032.69±106.64>(P< 0.05);those in the model group, high and middle dose drug group were markedly different from that in the normal control group(P< 0.05).CONCLUSION:Drug serum of highand middledose fufang biejia ruangan fang can inhibit the expression of αSMA in HSC.And its mechanism of curing hepatic fibrosis may be related with the inhibition of the conversion of αSMA from HSC by drug serum of fufang biejia ruangan fang.
