AIM:To further study the effects of platelet activating factor(PAF),inflammat ory cells and NO on cerebral ischemia-reperfusion injury in rat by given PAF r eceptor antagonist WEB2086. METHODS:One hundred Wister rats were randomly divided into two groups:① isch emia-reperfusion group(control group,n=50),which was subdivided into five grou ps with 10 rats in each group(pre-ischemia,ischemia for 2 hours,reperfusion fo r 2 hours,reperfusion for 12 hours and reperfusion for 24 hours);and② WEB 2086 treatment group(n=50),which consisted of five subgroups accorded with those in t he control group.Regional cerebral blood flow(rCBF),NO levels,myeloperoxidase(MP O) of leukocytes activity and infarction volume were determined respectively. RESULTS:WEB2086 significantly alleviated focal cerebral ischemia/reperfusion injury by reducing NO level,improving rCBF and suppressing MPO activity(from 3.9 4 ì mmol/L,85.8ì mmol/L,93.8 CONCLUSION:The protective effect of PAF receptor antagonist WEB2086 on cerebr al ischemia-reperfusion injury is related with NO.
