AIM: To study the effect of total glucosides of Centella asiatica on hyperplasia of mammary gland in rats. METHODS: SD rats
were randomly divided into seven groups( n =10): normal control group, model control group,Rupixiao positive group, tamoxifen positive group and three total glucosides of Centella asiatica groups with 1.5,3,6 mg·kg -1 . To adopt hyperplasia of mammary gland model, rats except normal control group were im in legs with estrogen (E_2) 0.025 mg·kg -1 ·d -1 for 25 d and Progesterone (P) 5 mg·kg -1 ·d -1 for 5 d to stimulate mammary gland. Rats in preventive treatment groups were administrated with total glucosides of Centella asiatica for 30 d while model started. Serum levels of E_2 and P were detected. Histological examination was performed. RESULTS: Total glucosides of Centella asiatica can inhibit hyperplasia of mammary gland growth induced by E_2 and P. Compared with model group serum level of E_2 in 3,6 mg·kg -1 total glucosides of Centella asiatica groups were decreased significantly ( P <0.05, P <0.01). Histological examination showed epithelial cell of acinus proliferation is obviously inhibited. CONCLUSION: Total glucosides of Centella asiatica have significant effect on hyperplasia of mammary gland in rats.
